Tissue culture of Dendrobium officinale refers to a propagation method in which, under sterile conditions, stem segments, shoot tips or seeds of Dendrobium officinale are taken as explants, inoculated onto an artificially prepared culture medium, and induced to differentiate to obtain complete seedlings. The seeds of Dendrobium officinale are tiny and difficult to germinate under natural conditions, and tissue culture technology precisely makes up for this shortcoming, becoming one of the main approaches for seedling breeding at present.

The tissue culture process roughly includes several stages: selection and disinfection of explants, primary induction, subculture proliferation, rooting culture, and hardening and transplanting. The disinfection stage is crucial; incomplete disinfection easily leads to contamination, while excessive disinfection harms the material, so repeated experimentation with the concentration of the agent and the treatment time is needed. In the proliferation stage, the ratio of the culture medium is adjusted to keep the shoots continuously dividing, after which they are transferred to rooting culture to promote root development.

Hardening before removal from the bottles is a step that cannot be ignored. Tissue-cultured seedlings have long been in a high-humidity, constant-temperature, sterile environment, and the survival rate of direct transplanting is often low. Generally, the bottles are first opened for a few days of hardening, then the residual culture medium on the roots is washed off, and the seedlings are moved into a loose, breathable substrate, maintaining relatively high air humidity and scattered light; after new roots grow, they are gradually shifted to routine management.

Compared with traditional methods such as division and cuttings, tissue culture propagation is fast, produces seedlings with a high degree of uniformity, and can also reduce the spread of disease, which is also of positive significance for protecting wild Dendrobium officinale resources. Although home growers find it difficult to carry out tissue culture operations, understanding its basic principles helps in identifying the source of seedlings when purchasing, and also allows for a better grasp of the key care points after transplanting.
