Detailed Explanation of the Method for Making Liquid Spawn of Edible Fungi
After installing the ventilation tube, use an oxygen supply machine to feed oxygen into the culture medium.
Place the culture medium on a shaker machine for shaking culture.
Liquid spawn of edible fungi has advantages such as a short production cycle, uniform and consistent spawn age, convenient inoculation, fast fermentation, and suitability for factory production. The production methods of liquid spawn of edible fungi can generally be divided into the electromagnetic stirring method, the shaker bottle method, the simple deep fermentation method, and the fermentation tank method. According to staff from the Edible Fungi Station of the Provincial Academy of Agricultural Sciences, the shaker bottle method is not only simple to inoculate and easy to manage, but its output also varies according to the machine's settings, and an investment of 2000-3000 yuan is enough.
To prepare the liquid culture medium, first boil potatoes until cooked but not mushy, then extract the supernatant, then prepare 3% glucose, 1% corn flour, 0.05% magnesium sulfate, and 0.1% potassium dihydrogen phosphate, and finally mix the potato supernatant with the above solution and water. The liquid culture medium is suitable for culturing many kinds of edible fungi spawn.
Shaker bottle shaking culture: After the liquid culture medium is prepared, put it into 500 ml Erlenmeyer flasks, with 100 ml in each flask, and add 1-15 small glass beads. Seal the flask mouth with a cotton plug and kraft paper, and sterilize for 30 minutes under a pressure of 1.5 kg/sq cm. Then add a piece of slant spawn about 2 sq cm, and culture statically at 23°C-25°C for 24 hours. When the hyphae germinate, place it on a reciprocating shaker for shaking culture, with a shaking frequency of 80-100 times per minute and an amplitude of 6-10 cm. The temperature of the shaker room is controlled at 24°C-25°C. The culture time varies depending on the type of fungus, generally about 5 days. Pour the shaken culture medium into a large container, and after volume expansion, install a ventilation tube under sterile conditions, feed in an appropriate amount of oxygen with an oxygen supply machine, and then culture for 48 hours. The standard for the end of culture: the culture liquid is clear and transparent, with a large number of small hyphal balls suspended in it, accompanied by the unique aroma of various mushrooms. The well-cultured liquid spawn should be stored in an incubator.
