Detailed Explanation of Key Points of Open Inoculation Technology for Shiitake Mushroom Logs

Open inoculation of shiitake mushrooms is a new technology summarized and innovated in recent years. Because it is easy to operate, does not require an inoculation box, and saves time and labor, it is generally welcomed by the majority of mushroom farmers. The technology is introduced as follows: 1. Selection of inoculation site ① Choose a room that is sheltered from the wind, clean, dry, well sealed, and with no sources of miscellaneous bacteria contamination within 50 meters nearby as the open inoculation site. ② The inoculation room must be cleaned and disinfected one day before the mushroom logs are moved in. 2. Mushroom log handling ① Under normal pressure sterilization, after maintaining at 100°C for 14 hours,

Open inoculation of shiitake mushrooms is a new technology summarized and innovated in recent years. Because it is easy to operate, does not require an inoculation box, and saves time and labor, it is generally welcomed by the majority of mushroom farmers. The technology is introduced as follows:

2. Mushroom log handling ① Under normal pressure sterilization, after maintaining at 100°C for 14 hours, move the mushroom logs without mechanical damage to the inoculation room and cover them with plastic film to prevent dust and miscellaneous bacteria spores from falling onto them. ② The mushroom logs are stacked in the middle of the inoculation room, and the stack height should not exceed 1.2 m. ③ Wait until the mushroom logs cool to below 25°C, disinfect with an aerosol disinfectant for 2 hours, and only then carry out inoculation.

3. Inoculation procedure ① Disinfect the spawn, inoculation tools, etc. with 75% alcohol and set aside for use. ② Wipe the inoculation site with a medicinal solution prepared from a sterilizing agent or carbendazim. ③ Punch holes at the wiped areas, punching 3 to 4 holes per mushroom log segment, with a hole depth of 4 cm to 5 cm, and do not punch through. ④ Break the production spawn into pieces and insert them into the holes. The spawn pieces and the mushroom log must be tightly joined, but not squeezed, to prevent miscellaneous bacteria from invading through the gaps. ⑤ When the temperature rises to 25°C, uncover the plastic film for ventilation and air exchange, and turn the stacks in time to prevent the spawn from being damaged by high temperature.

4. Precautions ① Inoculation must be carried out in low-temperature, clear, and dry weather. When inoculation is carried out at temperatures above 20°C, in rain and fog, or in windy conditions, there is a certain impact on the survival rate. ② The inoculation site must be clean, tidy, and hygienic. ③ The hands of personnel who come into contact with the spawn and the hole-punching machinery must be disinfected with 75% alcohol.
