Which type of inoculation tent works well for edible mushroom inoculation
1. Construction. The mosquito-net-style plastic inoculation tent is made from 3 m wide, 0.5 mm thick agricultural plastic film, and the specifications of the inoculation tent are 3 m long, 3 m wide and 3 m high. First cut a 13.5 m length of plastic film as the 3 m square enclosure wall of the inoculation tent, and take another 3 m length of film as the tent roof. Then use a plastic sealing machine or electric soldering iron to seal the two films along the edges, leave an overlapping joint line on one side of the tent as the door of the inoculation tent, and at the same time tie a rope at each of the 4 top corners of the inoculation tent for easy hanging.
2. Disinfection. Before use, first clean the inoculation site. Change the tent height to 2.5 m, and fold the remaining 0.5 m toward the inside of the tent. Then move the cooled mushroom sticks into the tent, stack them along the tent wall, and press the folded part of the plastic tent tightly and firmly. Leave an empty space in the middle of the tent for placing the inoculation table and for workers to move around. Before disinfection, put the spawn, cultivation bags and inoculation tools into the inoculation tent in advance, and after preparation is complete, close the inoculation tent for disinfection. Use aerosol disinfection boxes, 4-6 g per square meter, 50 g per box, and use 2 boxes for a 15 sq m inoculation tent. Light the disinfection boxes and disinfect in a closed state for 7-8 hours to achieve the sterilization effect. In addition, inoculation personnel must wear work clothes and work caps properly, maintain personal hygiene before entering the tent, trim their nails clean, and disinfect in a pre-prepared 5%-10% cresol soap aqueous solution; the arms should be soaked in 50-fold benzalkonium bromide solution for 5 minutes or wiped and disinfected with 75% alcohol cotton. When entering the tent, leave shoes outside the tent, and after entering, disinfect the inoculation tools with 75% alcohol cotton.
3. Inoculation. Use the double-bag inoculation method. In the double-bag method, after the bags are filled with a bagging machine, an outer bag is put on the outside; during inoculation, open the outer bag and make 5 or 4 holes, 2-3 cm deep, insert the spawn blocks into the holes, and then immediately tie the outer bag tightly; inoculation is complete. During inoculation, the inoculation tools should be wiped with 75% alcohol and sterilized by flame, sterilizing once after each use; the outer wall and mouth of the spawn bottle should be disinfected with 0.1% potassium permanganate aqueous solution. Place an alcohol lamp inside the inoculation tent; during inoculation, the mouth of the spawn bottle and the inoculation opening of the mushroom bag should be within the sterile zone of the alcohol lamp flame, and the movement of transferring spawn blocks should be fast. Each bottle of spawn packaged at 450 ml or 500 ml can inoculate 10-15 bags, with inoculation at both ends. The spawn blocks are best about the size of a broad bean. Inoculation operations should be carried out continuously: quickly untie the bag mouth, quickly inoculate, quickly tie the mouth, and quickly place them steadily; the tightness of the tied mouth should be appropriate. For good ventilation, a sterilized sponge block can be put in when tying the mouth. One inoculation tent holds about 1000 bags at a time, and inoculation should be finished within 4-5 hours, generally with a team of 3 people working together. After inoculation is complete, the mushroom sticks can be cultivated on site; the inoculated mushroom bags are stacked in a "#" shape, with 3 bags per row, placed 10 layers high. Keep the temperature in the greenhouse at 22-26°C, cover the greenhouse with straw curtains to prevent strong light, and remove the tent after the mycelium has grown to 5-10 cm. Some also transport the bags out and then spawn them separately. After each inoculation, clean the tools and miscellaneous items, do a good job of sanitation, carry out one fumigation disinfection with formaldehyde at 10-15 ml per cubic meter, and then inoculate again.
4. Advantages. Compared with an inoculation room (box), the inoculation tent has the advantages of low cost and high labor efficiency. Making a mosquito-net-style inoculation tent costs only more than 30 yuan, which is 1/5 of an inoculation box, and it is simple to make and can generally be used continuously for 3 years. An inoculation box inoculates about 25 mushroom sticks per hour, while an inoculation tent can reach more than 50 per hour, and it cultivates and spawns on site, with no need to use other things for sealing, saving labor.
