First, making the spawn: spawn is divided into mother spawn, stock spawn and planting spawn. Mother spawn is the spawn kept on a slant in test tubes; its preparation is relatively strict, and it is generally kept in research institutions or production units with the right conditions. Individuals or specialized households producing on a small scale can obtain it from these units.
Second, the stock spawn is made by expanding the mother spawn. Its raw materials are 75% hardwood sawdust, 23% wheat bran, 1% sugar, and 1% gypsum or calcium carbonate. Weigh the above raw materials in proportion according to the amount to be made, add water and stir evenly. The amount of water added is best when the hand is clenched tight and water appears between the fingers but does not drip. Then put the culture material into a canning jar; the canning jar should be washed clean beforehand and dried in the sun. While filling, press it down firmly with your hand, leaving a gap of 3-4 cm from the jar mouth; do not fill it level with the jar mouth. Then use a stick as thick as a finger to poke a hole in the material in the jar, going straight down to the bottom of the jar. Then use a cloth to wipe off the culture material attached to the jar and around the jar mouth, wrap the jar mouth with a layer of plastic film, tie it tightly with string or a rubber band, and sterilize it.
Third, there are two sterilization methods. One is to use a high-pressure sterilizer or pressure cooker and sterilize for half an hour to 1 hour. If there is no pressure cooker, use a steamer: place the filled canning jars one next to another in the pot, cover the pot tightly, and after steam rises, maintain it for 1-2 hours. To achieve thorough sterilization, it is best to let it stand overnight and then steam it for about another 1 hour.
Fourth, after sterilization, put the canning jars into an inoculation box; if there is no inoculation box, put them in a clean place. In front of the flame of an alcohol lamp, use an inoculation needle to transfer a piece of slant spawn the size of a fingernail into the hole in the middle of the canning jar. This operation must be done quickly to avoid contamination by stray microbes. After inoculation, cover the jar mouth again with plastic film, tie it well with string, and place it at 25-28 degrees C for constant-temperature culture. Generally, one slant spawn can inoculate 5-7 jars of stock spawn, and for each canning jar inoculated, the inoculation needle must be heated once over the alcohol lamp flame. After 20-30 days of culture, white mycelium can be seen filling the whole jar. This stage does not need light, and the air should be kept as fresh as possible.
Fifth, culture management: expand the stock spawn once more according to the stock spawn culture medium formula and operating method, and this is the planting spawn. Generally, one jar of stock spawn can inoculate 30-40 jars of planting spawn, and the culture temperature and time are basically the same as for stock spawn.
