How do you grow lion's mane mushroom indoors? Here are the cultivation methods

For indoor cultivation of lion's mane mushroom, you can refer to the following methods:

         1、Strain isolation and improved cultivation. Lion's mane mushroom cultivation first requires strain isolation. There are two methods of strain isolation: spore isolation and tissue isolation. Spore isolation involves collecting the spores released after the lion's mane mushroom matures under sterile conditions, as the material for cultivating mycelium. Tissue isolation involves taking fresh young fruiting bodies, placing them in an inoculation box, cutting open the mushroom body under sterile conditions, and taking a piece of tissue from the middle that is firm and of good texture, about the size of a grain of rice, and placing it onto a slanted culture medium in a test tube. The culture medium is made by mixing peeled potato, agar, and glucose, with a ratio of 200:20:20, adding 1000 g of water, adjusting the pH to 5.5-6, then filling into glass test tubes, not filling them completely, and sterilizing under high pressure to form a slanted surface. After inoculation, place the test tubes in a 25°C constant-temperature incubator for 3 days, and the inoculated block will begin to germinate mycelium; at 20-25 days, when the mycelium covers the surface of the culture medium, you can select well-grown cultures free of contaminating bacteria, and once the mycelium covers the slanted surface of the test tube culture medium, you can begin cultivating the spawn.

  2、Spawn cultivation. The substrate for spawn cultivation can use sawdust (free of aromatic oils or fungicides) or cottonseed hulls, etc. The ingredient ratios can be selected as follows:
(1) Sawdust 78%, wheat bran 20%, gypsum 1%, white sugar 1%;
(2) Cottonseed hulls 98%, gypsum 1%, white sugar 1%;
(3) Fresh distiller's grains 80%, wheat bran 10%, bran 9%, gypsum powder 1% (add inorganic salt nutrient solution, formula: ammonium sulfate 3%, potassium dihydrogen phosphate 1%, magnesium sulfate 0.1%, adjust to pH 4 with lactic acid.
(4). Add 60-65% water to the culture substrate and mix well; when squeezed by hand, there should be moisture between the fingers without dripping. Then, fill bottles to the shoulder and tamp firmly, and sterilize at 14.7 newtons pressure for 1 hour. After sterilization, use sterile technique to pick up a piece of slanted spawn and put it into the culture medium bottle, bring the temperature to 20-25°C and the relative air humidity to 60-65%, and plug with a cotton plug. After about 30-40 days, the mycelium covers the substrate and forms a mycelial membrane. When the fruiting body primordia on the mycelial membrane approach the cotton plug, remove the cotton plug.

  3、Fruiting body cultivation. Open cultivation of lion's mane mushroom involves sterilizing broadleaf tree sawdust under high pressure, or boiling it in a pot for 20 minutes, removing and draining the water, and after cooling, directly mixing in about 40% of the bottle spawn; after mixing, put it into plastic bags or wrap it with film. Fit the bag opening or tied opening with a cotton plug for ventilation, and place it at a low temperature below 10°C for cultivation. After it grows into a mycelial block, remove the film and continue cultivating at natural temperature. After a few months, lion's mane mushrooms will grow. This method gives a yield similar to bottle cultivation, but saves spawn and bottles, and allows for larger-scale production. For bottle-cultivated lion's mane mushroom, after pulling out the cotton plug, keep the room temperature at 18-20°C. If the temperature is too high, close doors and windows at noon and ventilate in the early morning and evening when the temperature is lower; if the temperature is low, ventilate at noon. Keep indoor humidity at 85-95%; if too low, sprinkle water on the floor to keep it constantly moist, and also spray the air 3-4 times a day. After half a month, fruiting bodies will form. Compared with sawdust and cottonseed hulls as substrate, cottonseed hulls have the characteristics of faster mycelial growth, fewer contaminating bacteria, and earlier fruiting body formation, forming fruiting bodies about half a month earlier than sawdust.

  4、Harvesting. When the fruiting body begins to shrivel and age and stops growing and developing, and the surface turns from white to brown, it can be harvested and dried in the sun. If for consumption, it can be harvested before the spores are released, which avoids the bitter taste of the spores. Under the same management conditions as above, bottle-cultivated lion's mane mushroom can harvest three per bottle, and the remaining mycelium can be used for medicine.
