Conditions and Equipment for Plant Tissue Culture

Plant tissue culture involves culturing plant materials under strictly sterile conditions, so certain laboratory conditions are required. Regardless of its level, a culture room generally consists of the following parts: 1. Washing room: for washing and storing the large quantity of glassware used in tissue culture; it requires water supply and drainage facilities. 2. Medium preparation room (or chemistry laboratory): the place for preparing culture media. It needs to be equipped with various chemical reagents, various graduated cylinders, pipettes, test tubes and other glassware; a balance for weighing; a refrigerator for storing stock solutions, etc. 3. Autoclaving room: after the culture medium is prepared, it needs to be autoclaved to kill contaminating microorganisms, so an autoclave is required, and the electricity and heat supply must be safe and convenient. 4. Inoculation room: an important operating room for isolating and inoculating plant materials and transferring cultured materials. It should be flat, tidy and clean, and easy to clean and sterilize. If conditions permit, a clean bench should be purchased; a simple sterile inoculation box can also be used instead. 5. Culture room: the place where the inoculated materials are cultured. Its size depends on need, and culture racks are usually placed in it. The room temperature should not be too high or too low; if conditions permit, an air conditioner should be installed, and fluorescent lamps should be installed to provide light. Natural light can also be used to reduce energy consumption. According to the arrangement of the above parts, the main equipment for plant tissue culture includes: glassware, a refrigerator, an ordinary balance and an analytical balance, an autoclave, a clean bench or inoculation box, culture racks, etc. (Wang Huaiyu)
