How are amoebas cultured? A detailed introduction to the culturing method
How to culture amoebas?
Amoebas like to live in pools with relatively clear water or in shallow water with slow-flowing water and abundant algae. Sometimes they attach to decaying plants submerged in water or on the muddy bottom, or float on foam on the water surface. Take mud from the bottom surface of the pond or decaying organic matter, collect it together with water, put it in a bottle and let it stand for about 24 hours. Use this as the source when culturing. The optimum temperature for amoebas is 18°C~22°C, and they are easiest to collect in spring and autumn. Amoebas are relatively large in body size, and a white spot is visible to the naked eye. Therefore, advantage can be taken of the characteristic that when well fed and suddenly shaken, they will firmly attach to objects, so they can be separated out and used as a seed source. The method is to take culture liquid containing amoebas and drop it onto a glass slide; the amoebas will firmly attach to the glass slide, then slowly rinse the culture liquid off the glass slide with cool water for about 10 seconds. In this way, make several slides as a seed source, put them together with the glass slides into culture liquid, and after several days of culturing, a large number of relatively pure amoebas can be obtained.
