I. Preparation before artificial insemination in pigs

1. Before semen collection, the semen collection cup needs to be placed in a constant-temperature cabinet at 35-37°C for insulation, so as to prevent sperm from being affected by temperature differences, and direct sunlight should also be avoided on the semen.

2. Boars generally used for artificial insemination do not need to undergo natural mating, to avoid affecting the boar's libido and mood during semen collection. In order to ensure the boar's libido and high-quality semen, 65% are generally replaced each year.

3. The collected semen must be strictly examined. After semen collection is completed, the semen should be immediately placed in a 37°C constant-temperature cabinet, and strict quality and artificial insemination checks should be carried out as soon as possible.

II. Observe the color

1. Generally, normal semen is milky white or grayish white. Semen with high concentration is usually milky white with a yellowish-white tint, while semen with low concentration is grayish white, watery, or even transparent.

2. If the semen is reddish-brown, it may be mixed with blood. If it is yellow and has a foul odor, it may be mixed with urine or inflammatory secretions. Such semen cannot be used, and the boar should also be treated symptomatically in a timely manner.

III. Odor

Normal semen has no special odor. Semen with a urine odor, ammonia odor, or other strange odors cannot be used.

IV. PH value

Generally, the lower the PH value of semen, the greater the sperm concentration. Normal semen is neutral to weakly alkaline, at about 7.0-7.8.

V. Sperm motility and abnormality rate

1. Sperm motility and abnormality rate can only be observed under a 200-400x microscope. If forward-moving sperm account for less than 60% of the total, it cannot be used.

2. Then use an electronic calculator to measure sperm density, calculate the number of sperm contained in the collected semen, and calculate the dilution factor according to each insemination dose of at least 3 billion effective sperm. Generally, it can be diluted 10-15 times.

VI. Semen dilution

1. First prepare professional commercial diluent and self-made distilled water. The temperature of the diluent and the temperature of the semen to be diluted need to be consistent, and the temperature difference cannot exceed 1°C.

2. Semen can be diluted according to the dilution factor. An automatic mixer and glass rod can be used for thorough mixing and stirring, then use a 100 ml pointed plastic bottle, and non-toxic and harmless plastic bags are needed for sub-packaging, labeled with the boar number and semen collection time for differentiation.

3. Finally, store the diluted semen. After placing it at room temperature of 25°C for 1-2 hours, put it into a constant-temperature cabinet for preservation. It can be turned once every 6 hours or 12 hours, and can be preserved for 5-6 days. Generally, it is required to be used up within 3 days, otherwise it will affect the fine insemination effect.
